  • Male Wistar rats were conditioned to consume a glucose solution.
  • Hypothalamic ventromedial (VMH) and lateral (LH) regions were analyzed for monoamine levels and metabolites.
  • Serum insulin and glucose levels were measured.

  • Conditioned rats exhibited significantly higher serum insulin levels compared to controls, with unchanged glucose levels.
  • Increased norepinephrine, dihydroxyphenylethylene glycol (DHPG), and serotonin (5-hydroxytryptamine, 5-HT) were observed in the LH.
  • Elevated 5-HT, 5-hydroxyindoleacetic acid, and 3,4-dihydroxyphenylacetic acid (DOPAC) were found in the VMH.
  • LH 5-HT and VMH DOPAC levels strongly correlated with serum insulin.

Conclusions:

  • Hypothalamic monoamine systems, specifically norepinephrine and serotonin in the LH and serotonin and dopamine metabolites in the VMH, are implicated in cephalic phase insulin release.
  • These findings define the changes in specific hypothalamic monoamines associated with anticipatory insulin secretion in normal animals.
