Ribosome Profiling02:24

Ribosome Profiling

Ribosome profiling or ribo-sequencing is a deep sequencing technique that produces a snapshot of active translation in a cell. It selectively sequences the mRNAs protected by ribosomes to get an insight into a cell’s translation landscape at any given point in time.
Applications of ribosome profiling
Ribosome profiling has many applications, including in vivo monitoring of translation inside a particular organ or tissue type and quantifying new protein synthesis levels.
The technique helps...

Area of Science:

  • Avian biology
  • Biomaterials science
  • Molecular genetics

Background:

  • Avian eggshell membranes are crucial for reproduction and have industrial/biomedical potential.
  • Their insoluble nature makes formation and protein composition challenging to study.
  • Understanding these membranes is key to improving eggshell quality.

Purpose of the Study:

  • To identify genes encoding eggshell membrane proteins.
  • To characterize proteins responsible for the structural integrity of eggshell membranes.
  • To analyze the transcriptome of the white isthmus for key gene expression.

Main Methods:

  • Utilized the Del-Mar 14 K chicken microarray.
  • Compared gene expression in the white isthmus (WI) with magnum (Ma) and uterine (Ut) oviduct segments.
  • Analyzed over-expressed transcripts to identify specific gene IDs.

Main Results:

  • Identified 107 Gallus gallus gene IDs with over-expressed transcripts in the WI.
  • Found significant upregulation of structural proteins: collagen X (COL10A1), fibrillin-1 (FBN1), and cysteine-rich eggshell membrane protein (CREMP).
  • Detected over-expression of genes involved in collagen processing (LOXL1-3, P4HA2, P4HB), isomerases (PPIC), and disulfide cross-linking (QSOX1, TXN).

Conclusions:

  • Identified genes critical for collagen processing and disulfide cross-linking in eggshell membrane formation.
  • Gained new insights into eggshell membrane structure and formation mechanisms.
  • Findings support developing strategies to enhance eggshell quality and food safety.
