    Benign breast conditions show increased plasminogen and angiostatin levels, except in proliferative fibroadenomas. Angiostatin may play a key role in non-cancerous breast diseases, suggesting potential diagnostic and therapeutic implications.

    Background:

    • Benign breast tissue has low angiogenic capacity.
    • Angiogenesis activation in pre-malignant lesions may drive breast cancer progression.
    • Molecular mechanisms of angiogenesis in non-cancerous breast pathologies are poorly understood.

    Purpose of the Study:

    • To quantify plasminogen and angiostatin levels in benign breast conditions.
    • To investigate the role of plasminogen fragments in mastopathy, breast cysts, and fibroadenomas.
    • To explore the link between matrix metalloproteinase-9 and angiostatin in benign breast lesions.

    Main Methods:

    • Immunoblotting and densitometric scanning for plasminogen and angiostatin quantification.
    • Gelatine zymography for matrix metalloproteinase-9 activity detection.
    • Comparative analysis of protein levels in healthy breast tissue versus various benign pathologies.

    Main Results:

    • Significant increases in plasminogen (3.5-4.7 fold) and angiostatin (3.7-6.7 fold) were observed in fibrocystic changes, cysts, and non-proliferative fibroadenomas compared to controls.
    • Matrix metalloproteinase-9 activation may contribute to increased angiostatin in fibrocystic and cyst masses.
    • Proliferative fibroadenomas showed decreased plasminogen (3.8 fold) and angiostatin (5.3 fold) compared to dormant fibroadenomas.

    Conclusions:

    • Angiostatin, an inhibitor of vessel growth and inflammation, appears crucial in the pathophysiology of non-cancerous breast diseases.
    • Altered plasminogen and angiostatin levels may serve as biomarkers for benign breast conditions.
    • Further research is warranted to explore the diagnostic and therapeutic potential of these proteins in benign breast pathologies.
