Objective:
In this study, we screened the different human osteosarcoma cell line MG-63 miRNAs after the treatment of curcumin and explored the effects of curcumin on MG-63 cells and its mechanism.
Methods:
Affemitrix miRNA chip was used to detect the changes of miRNA expression profile in MG-63 cells before and after curcumin treatment, and screen different expression of miRNAs. The target gene of miRNA was analyzed by bioinformatics. The expression levels of miRNA-138 target genes Smad4, NFκB p65 and cyclin D3 were detected. MTT and Transwell Cell invasion assays were used to observe the effects of curcumin on MG-63 cells.
Results:
Curcumin could significantly inhibit the proliferation of MG-63 cells and the expression levels of miRNA-138 target genes Smad4, NFκB p65 and cyclin D3 in MG-63 cells (P<0.05); overexpression of hsa-miR-138 down-regulated the expression levels of Smad4, NFκB p65 and cyclin D3 compared with the treatment of curcumin, while inhibition of hsa-miR-138 up-regulated the expression levels of Smad4, NFκB p65 and cyclin D3.
Conclusions:
Curcumin could increase the expression of hsa-miR-138, hsa-miR-138 inhibited cell proliferation and invasive ability by inhibition of its target genes.
Insights
Curcumin treatment increases hsa-miR-138 expression in osteosarcoma cells, inhibiting proliferation and invasion by targeting Smad4, NFκB p65, and cyclin D3.
Area of Science:
- Oncology
- Molecular Biology
- Pharmacology
Background:
- Osteosarcoma is a primary bone malignancy with limited treatment options.
- Curcumin, a compound from turmeric, exhibits potential anti-cancer properties.
- MicroRNAs (miRNAs) play crucial roles in cancer development and progression.
Purpose of the Study:
- To investigate the effects of curcumin on human osteosarcoma cell line MG-63.
- To identify differentially expressed miRNAs in MG-63 cells following curcumin treatment.
- To elucidate the mechanism of curcumin's action on MG-63 cells.
Main Methods:
- miRNA expression profiling using Affymetrix miRNA chips.
- Bioinformatic analysis to predict miRNA targets.
- Validation of target gene expression (Smad4, NFκB p65, cyclin D3) via quantitative methods.
- Assessment of cell proliferation (MTT assay) and invasion (Transwell assay).
Main Results:
- Curcumin significantly inhibited MG-63 cell proliferation and invasion.
- Curcumin treatment led to increased expression of hsa-miR-138.
- Overexpression of hsa-miR-138 downregulated Smad4, NFκB p65, and cyclin D3; inhibition of hsa-miR-138 upregulated these targets.
Conclusions:
- Curcumin enhances hsa-miR-138 expression in osteosarcoma cells.
- Hsa-miR-138 mediates curcumin's inhibitory effects on cell proliferation and invasion.
- Targeting Smad4, NFκB p65, and cyclin D3 is a key mechanism for curcumin's anti-osteosarcoma activity.
