    Summary

We investigated orientation recovery in single-molecule imaging using diffusion maps. A modified scattering image approach improves orientation recovery for realistic molecules, enhancing imaging analysis.

    Area of Science:

    • Structural biology
    • Computational imaging
    • Data analysis

    Background:

    • Single-molecule imaging techniques are crucial for understanding molecular structures.
    • Coherent diffractive imaging (CDI) offers high-resolution structural information.
    • Orientation recovery is a key challenge in single-molecule CDI.

    Purpose of the Study:

    • To explore orientation recovery in single-molecule CDI using diffusion maps.
    • To identify limitations of diffusion map-based orientation recovery for realistic molecules.
    • To propose a modified approach for improved orientation recovery.

    Main Methods:

    • Application of diffusion map algorithm to approximate the Laplace-Beltrami operator.
    • Diagonalization of the operator using a metric for Euler angle mapping.
