Background:

  • The bovine oviduct plays a critical role in early embryonic development and maternal recognition of pregnancy.
  • Understanding the transcriptomic landscape of the oviduct is essential for improving reproductive efficiency in cattle.

Purpose of the Study:

  • To compare the oviductal isthmus transcriptome between pregnant and cyclic heifers.
  • To investigate spatial transcriptomic differences between the oviductal isthmus and ampulla in pregnant heifers.

Main Methods:

  • Microarray analysis of epithelial cells from the oviductal ampulla and isthmus of synchronized beef heifers on Day 3 post-oestrus.
  • Heifers were categorized as cyclic (non-bred) or pregnant (artificially inseminated).
  • Gene ontology analysis to identify overrepresented biological processes.

Main Results:

  • No significant effect of corpus luteum proximity on the isthmus transcriptome, regardless of pregnancy status.
  • 2287 differentially expressed genes (P<0.01) between the ampulla and isthmus in pregnant heifers.
  • Isthmus: enriched for synthesis (nitrogen, lipids, nucleotides, steroids, cholesterol), transport, cell cycle, and apoptosis. Ampulla: enriched for cell motion, DNA repair, ion homeostasis, carbohydrate biosynthesis, and ciliary function.

Conclusions:

  • Significant transcriptomic divergence exists between the bovine oviductal ampulla and isthmus by Day 3 of pregnancy.
  • These regional differences suggest specialized roles in supporting early embryonic development and/or transport.
