Human mesenchymal stem cells (MSCs) and fibroblasts share similar characteristics, making them difficult to distinguish based on current definitions. This study found no unique immunophenotypic properties to differentiate these cell types.
Area of Science:
- Cell Biology
- Immunology
- Stem Cell Research
Background:
- Human mesenchymal stem cells (MSCs) exhibit fibroblast-like morphology and differentiation potential.
- Distinguishing MSCs from fibroblasts based on phenotype remains challenging.
- Existing research highlights shared characteristics between MSCs and fibroblasts.
Purpose of the Study:
- To compare the phenotypic and immunological characteristics of human MSCs and fibroblasts.
- To identify distinguishing properties between MSCs and fibroblasts.
- To evaluate the potential for immunomodulation in both cell types.
Main Methods:
- Utilized established human fibroblast strains from multiple tissue sources.
- Employed human MSC strains from various tissue origins.
- Conducted comparative analysis of phenotypic and immunological markers.
Main Results:
- Fibroblast strains displayed morphology, cell surface markers, and differentiation capabilities similar to MSCs.
- Both cell types demonstrated T cell proliferation suppression and macrophage immunomodulation.
- Extracellular matrix deposition and FSP1 expression patterns were comparable between fibroblasts and MSCs.
Conclusions:
- No distinct immunophenotypic properties were identified to differentiate human MSCs from fibroblasts.
- Current definitions do not provide a clear distinction between these cell types.
- Further research may be needed to establish definitive markers for MSCs and fibroblasts.

Adult stem cells
Adult stem cells are tissue-specific; hence, they divide to develop the tissue from which they originate. One type of adult stem cell is the epithelial stem cell, which gives rise to the keratinocytes in the multiple layers of epithelial cells in the epidermis of the skin. Adult bone marrow has three distinct types of stem cells:...
