Area of Science:

  • Dermatology
  • Biomaterials Science
  • Regenerative Medicine

Background:

  • Forehead wrinkles stem from frontalis muscle contraction and skin aging.
  • Current treatments include hyaluronic acid fillers and botulinum toxin.
  • Polydioxanone (PDO) threads are also used for wrinkle correction.

Purpose of the Study:

  • To evaluate the efficacy and safety of a multi-polydioxanone (PDO) scaffold for treating forehead wrinkles.
  • To assess the scaffold's performance in animal models and human skin.

Main Methods:

  • A stent-shaped multi-PDO scaffold was implanted in rat and mini-pig skin.
  • The scaffold was also applied to forehead wrinkles in three human volunteers.
  • Histological and photographic evaluations were conducted.

Main Results:

  • Histological analysis at 12 weeks demonstrated new collagen synthesis (de novo collagen synthesis).
  • Clinical photographic evaluations corroborated the histological findings.
  • The treatment showed positive results in both animal models and human subjects.

Conclusions:

  • Stent-shaped multi-PDO scaffolds represent a potentially effective and safe treatment for reducing forehead wrinkles.
  • This novel scaffold may offer an alternative therapeutic option for aesthetic concerns related to aging skin.
