Flow Cytometry01:23

Flow Cytometry

17.1K
The development of flow cytometry techniques began in 1934 with initial attempts by Andrew Moldavan, a bacteriologist who counted the cells in a flowing capillary system. Moldavan pumped cells through a capillary tube focused under a microscope for visualization. The invention of photometry allowed the measurement of differentially-stained cells, and Louis Kamentsky developed the first multiparameter flow cytometer in 1965 to identify and count the cancer cells in cervical tissue specimens.

Area of Science:

  • Plant genomics
  • Cytogenetics
  • Molecular biology

Background:

  • Nuclear genomes in plants are complex and challenging to map and sequence.
  • Isolating single chromosomes is crucial for various plant research fields.

Purpose of the Study:

  • To describe a detailed protocol for preparing liquid suspensions of intact mitotic metaphase chromosomes.
  • To enable flow cytometric analysis and sorting of these chromosomes.

Main Methods:

  • Preparation of intact mitotic metaphase chromosomes from plant tissues.
  • Flow cytometric analysis for chromosome characterization.
  • Flow sorting for isolation of pure chromosome fractions.

Main Results:

  • Successful isolation of intact mitotic metaphase chromosomes in liquid suspension.
  • Demonstration of flow cytometric analysis and sorting capabilities for plant chromosomes.

Conclusions:

  • Flow cytometry is the premier method for obtaining pure chromosome fractions from plants.
  • This protocol facilitates downstream applications like physical mapping and sequencing.
