  • Sucrose intake test assessed antidepressant effects; 5-HT1A receptor binding assay (using 8-OH-DPAT) measured receptor changes in the hippocampus, frontal cortex, and hypothalamus.
  • Main Results:

    • N. s. significantly reversed CMS-induced decreases in sucrose intake.
    • Both N. s. and H. p. reversed CMS-induced decreases in 5-HT1A receptor binding in the hippocampus and frontal cortex.
    • N. s. and H. p. showed greater increases in frontal cortex 5-HT1A binding than fluoxetine; all treatments normalized binding in the hypothalamus.

    Conclusions:

    • Nelumbinis Semen exhibits antidepressant properties by enhancing 5-HT1A receptor binding.
    • The mechanism of N. s. is comparable to H. p. in modulating 5-HT1A receptor binding, differing from fluoxetine.
    • N. s. represents a potential therapeutic agent for depression through its effects on the serotonin system.
