When T cells with CD4 markers are activated, they give rise to two types of effector cells: helper T cells and regulatory T cells. Meanwhile, T cells with CD8 markers differentiate into effector cytotoxic T cells. The differentiation of CD4 T cells into helper T cell subsets, such as Th1, Th2, and Th17 cells, is dependent on the antigen type, antigen-presenting cell, and regulatory cytokines.

Background:

  • Dendritic cells (DCs) are key antigen-presenting cells.

Purpose of the Study:

  • To investigate histamine's effect on murine DC antigen uptake and activation.
  • To identify histamine receptor subtypes involved.

Main Methods:

  • Murine splenic and lung DCs were isolated and stimulated with histamine.
  • Antigen uptake and co-stimulatory molecule expression (CD80, CD86, ICOS-L) were measured by flow cytometry.
  • Histamine receptor antagonists (H1R, H2R, H4R) were used to determine receptor involvement.

Main Results:

  • Histamine accelerated antigen uptake in DCs via H1R, H2R, and H4R.
  • Histamine enhanced CD86 and ICOS-L expression, but not CD80.
  • Blocking H1R, H2R, and H4R reduced histamine-induced co-stimulatory molecule expression.

Conclusions:

  • Histamine regulates the immunological synapse.
  • Histamine stimulates DC antigen uptake and activation through H1R, H2R, and H4R.
