Multiple Comparison Tests01:13

Multiple Comparison Tests

4.5K
Multiple comparison test, abbreviated as MCT, is a post hoc analysis generally performed after comparing multiple samples with one or more tests. An MCT will help identify a significantly different sample among multiple samples or a factor among multiple factors.
It would be easy to compare two samples using a significance alpha level of 0.05. In other words, there is only one sample pair to be compared. However, it would be difficult to identify a significantly different sample if the number...

Sample Preparation for Analysis: Advanced Techniques

1.6K
Accurate analysis of complex samples often requires advanced preparation techniques to achieve reliable and reproducible results. Samples containing inorganic or organic materials can be challenging to dissolve or decompose effectively. Standard sample preparation methods include acid digestion, fusion, dry ashing, and wet digestion.
Acid digestion with strong acids is commonly used to dissolve inorganic materials that are insoluble (do not dissolve) in water. This method can be useful for...

Area of Science:

  • Reproductive biology
  • Genomics
  • Epigenetics

Background:

  • Gene expression analysis is crucial for understanding human pregnancy development.
  • Chorionic villi samples (CVS) offer a window into ongoing pregnancies but require optimized processing.
  • Current methods for multi-platform testing of CVS are not well-established.

Purpose of the Study:

  • To establish optimal methods for storing and processing chorionic villi samples (CVS) for comprehensive genetic, epigenetic, and gene expression analyses.
  • To evaluate the utility of RNAlater preservation versus flash-freezing for CVS.
  • To develop techniques for utilizing small CVS amounts for multiple molecular assays.

Main Methods:

  • Chorionic villi samples were preserved using either flash-freezing or RNAlater.
  • Modified DNA and RNA isolation kits were used to extract nucleic acids from small sample amounts (2-5 mg).
  • RNA quality and yield were assessed, and RNA-sequencing (RNA-seq) libraries were prepared and analyzed.

Main Results:

  • RNAlater preserved samples yielded significantly higher RNA quality and quantity compared to flash-frozen samples.
  • RNA-seq analysis using RNAlater samples demonstrated consistent biological variation regardless of starting RNA amount (200 vs 800 ng).
  • DNA isolation from RNAlater samples improved with heated elution buffers, and purification was unnecessary for methylation profiling.

Conclusions:

  • RNAlater is the superior method for preserving chorionic villi samples for molecular studies.
  • Optimized techniques allow for integrated genetic, epigenetic, and gene expression profiling from single, small CVS samples.
  • These advancements facilitate the development of prenatal diagnostics and therapeutics via a systems biology approach.
