Area of Science:

  • Biochemistry
  • Spectroscopy
  • Melanogenesis research

Background:

  • Melanin quantification is crucial for understanding pigmentation disorders and melanoma.
  • Absorption spectroscopy (405 nm) is the current standard but has significant limitations.

Purpose of the Study:

  • To introduce and validate fluorescence spectroscopy as a superior method for melanin quantification.
  • To address the inaccuracies and limitations of absorption spectroscopy in melanin assessment.

Main Methods:

  • Implementation of fluorescence spectroscopy for in vitro melanin quantification.
  • Comparison of fluorescence and absorption spectroscopy in cell-based assays.
  • Application of fluorescence spectroscopy to complex biological matrices (zebrafish embryos, human hair).

Main Results:

  • Absorption spectroscopy overestimates melanin content and cannot differentiate control from melanogenic cells.
  • Fluorescence spectroscopy demonstrates high specificity and accuracy in detecting melanin variations.
  • Successful application of fluorescence spectroscopy in complex biological samples.

Conclusions:

  • Fluorescence spectroscopy is the optimal method for accurate and specific melanin quantification.
  • This technique overcomes the limitations of absorption spectroscopy for melanin assessment.
  • The method holds promise for diverse research applications, including in vivo studies.
