Skin Diseases and Disorders

6.0K
Skin is the first line of defense and encounters a variety of microbes. Some pathogenic strains are often the cause of a broad range of infections of the skin and other body systems. These conditions can affect people of all ages and may have different causes, including genetic factors, infections, autoimmune reactions, environmental factors, and lifestyle choices.
Gram-positive Staphylococcus spp. and Streptococcus spp. are responsible for many of the most common skin infections. However, many...

Inflammatory Response01:28

Inflammatory Response

18.0K
An inflammatory response is a localized, nonspecific immune reaction that occurs when a tissue is injured. It is characterized by redness, swelling, heat, and pain, which are commonly called the cardinal signs and symptoms of inflammation. Inflammation can sometimes result in a loss of function.
Inflammation can be triggered by various stimuli, such as impact, abrasion, chemical irritation, infections, and extreme hot or cold temperatures. These can damage cells and connective tissue fibers,...

Immunofluorescence Microscopy01:12

Immunofluorescence Microscopy

14.4K
A fluorescence microscope uses fluorescent chromophores called fluorochromes, which can absorb energy from a light source and then emit this energy as visible light. Fluorochromes include naturally fluorescent substances (such as chlorophylls) and fluorescent stains that are added to the specimen to create contrast. Dyes such as Texas red and FITC are examples of fluorochromes. Other examples include the nucleic acid dyes 4’,6’-diamidino-2-phenylindole (DAPI), and acridine orange.

The JAK-STAT Signaling Pathway01:20

The JAK-STAT Signaling Pathway

13.6K
Several cytokine receptors have tightly bound Janus kinase or JAK proteins attached at their cytosolic tail. Small signaling molecules such as cytokines, growth hormones, or prolactins bind to the cytokine receptors and initiate their dimerization. The dimerization brings the cytosolic JAKs together that trans-phosphorylate and activates each other. The activated JAKs now phosphorylate cytosolic tails of the cytokine receptors, which serve as binding sites for adaptor proteins such as  SH2...

Inflammatory Response I: Vascular and Cellular01:30

Inflammatory Response I: Vascular and Cellular

17.8K
The inflammatory response is the body's defense against infection, injury, or irritation from bacteria, trauma, toxins, or heat. Inflammation helps locate and destroy pathogens and remove damaged tissue elements to heal the body. During this initial phase, fluid, blood products, and nutrients migrate to the injured area, resulting in redness, heat, swelling, ache, and loss of function. Moreover, signs of systemic inflammation include fever, increased WBC count, malaise, anorexia, nausea,...

Summary

Inflammasomes, key innate immune sensors, are implicated in numerous skin diseases, from autoinflammatory syndromes to infections and malignancies. Targeting inflammasome pathways offers promising therapeutic strategies for various dermatological conditions.

Area of Science:

  • Immunology
  • Dermatology
  • Molecular Biology

Background:

  • Inflammasomes are intracellular multiprotein complexes central to innate immunity.
  • Dysregulation of inflammasomes is increasingly linked to a spectrum of human diseases.
  • Autoinflammatory diseases involve innate immune cell activation leading to tissue damage.

Purpose of the Study:

  • To review the literature on the role of inflammasomes in various skin diseases.
  • To highlight inflammasome involvement in monogenic autoinflammatory syndromes with skin manifestations.
  • To explore inflammasome participation in other dermatological conditions, including infections, malignancies, and autoimmune diseases.

Main Methods:

  • Literature review of scientific articles.
  • Analysis of studies investigating inflammasome components and activation in skin pathologies.
