Ectonucleotide pyrophosphate phosphodiesterase-1 (ENPP1) is elevated in gestational diabetes mellitus (GDM) adipose tissue, correlating with insulin resistance. Overexpression of ENPP1 in mice impairs glucose tolerance, suggesting a causative role in GDM.
Area of Science:
- Endocrinology and Metabolism
- Molecular Biology
- Reproductive Medicine
Background:
- Gestational diabetes mellitus (GDM) is a condition marked by insulin resistance and disrupted glucose-lipid metabolism during pregnancy.
- Ectonucleotide pyrophosphate phosphodiesterase-1 (ENPP1) is implicated in adipocyte insulin resistance.
- This study investigates ENPP1's role as a potential determinant of GDM.
Purpose of the Study:
- To examine ENPP1 expression in the adipose tissue (AT) of obese pregnant women with and without GDM.
- To assess glucose tolerance in pregnant mice with AT-specific overexpression of ENPP1.
Main Methods:
- Collected AT biopsies and plasma from obese pregnant women (GDM and non-GDM) and nonpregnant controls.
- Measured key insulin signaling and glucose-lipid metabolism molecules in AT.
- Assessed plasma glucose and insulin, calculated HOMA-IR, and performed intraperitoneal glucose tolerance tests in transgenic mice.
Main Results:
- Obese GDM patients exhibited significantly higher AT ENPP1 expression compared to obese non-GDM patients and controls.
- ENPP1 expression correlated negatively with glucose transporter 4 (GLUT4) and positively with insulin receptor substrate-1 (IRS-1) serine phosphorylation.
- Elevated AT ENPP1 levels were positively correlated with HOMA-IR, and pregnant ENPP1 transgenic mice showed impaired glucose tolerance.
Conclusions:
- Findings suggest a causative link between ENPP1 and metabolic dysregulation in GDM.
- ENPP1 may mediate insulin resistance and hyperglycemia in GDM by affecting insulin signaling, glucose uptake, and lipid metabolism in adipose tissue.
