Endothelial Colony Forming Cells (ECFCs) in murine AKI - implications for future cell-based therapies

Endothelial Colony Forming Cells (ECFCs) transiently improve kidney function and reduce fibrosis post-ischemia in mice. However, ECFCs do not prevent capillary loss or modulate p62 levels, showing less efficacy than early EPCs in long-term damage prevention.

Purpose of the Study:
- To evaluate the effects of Endothelial Colony Forming Cells (ECFCs) on post-ischemic kidney function over several weeks.
- To analyze ECFCs' impact on endothelial-to-mesenchymal transition (EndoMT), peritubular capillary density (PTCD), and endothelial markers (aT, p62).
Main Methods:
- Mice underwent bilateral renal pedicle clamping (35 or 45 min).
- Syngeneic ECFCs were intravenously injected post-ischemia.
- Kidney function, fibrosis, PTCD, EndoMT, aT, and p62 were assessed at 1, 4, and 6 weeks.
Main Results:
- ECFCs improved kidney function only at week 1.
- Ischemia-induced fibrosis was reduced by ECFCs, but PTCD loss was unaffected.
- ECFCs partially reduced EndoMT in one group and aggravated endothelial aT decline; p62 levels were not modulated.
Conclusions:
- Single-dose ECFCs reduce long-term fibrosis but offer only transient functional improvement.
- ECFCs differ from eEPCs, not preventing capillary loss or altering p62.
- ECFC therapy is less effective than eEPC therapy in mitigating ischemia-induced kidney damage.
