    Area of Science:

    • Immunology
    • Cell Biology
    • Biochemistry

    Background:

    • Thy-1 antigen is a cell surface glycoprotein found on mouse thymocytes.
    • Understanding Thy-1's biosynthesis and maturation is crucial for immunology and cell biology research.

    Purpose of the Study:

    • To investigate the biosynthesis and maturation pathway of mouse Thy-1 antigen.
    • To characterize the structural modifications and cellular localization of Thy-1 during its development.

    Main Methods:

    • Pulse-chase labeling with [35S]methionine and [35S]cysteine.
    • Analysis of molecular weight changes using SDS-PAGE.
    • Enzymatic digestion with endo-beta-N-acetylglucosaminidase H and F.
    • Tunicamycin treatment to assess glycosylation.
    • Trypsin digestion to determine membrane association.

    Main Results:

    • The earliest Thy-1 form (26,500 Da) matures into a 25,000 Da polypeptide through glycan trimming.
    • Mature Thy-1 appears at the cell surface within 15 minutes, retaining high-mannose N-linked glycans.
    • Endo-F digestion reveals a common precursor (13,500 Da) for both early and late forms, indicating no O-linked glycosylation.
    • Thy-1 lacks a cytoplasmic domain, as evidenced by its resistance to trypsin digestion when membrane-associated.

    Conclusions:

    • Mouse Thy-1 undergoes significant post-translational modifications, including N-linked glycosylation and glycan trimming, during its maturation.
    • The mature Thy-1 molecule is a cell surface protein with no exposed cytoplasmic portion.
    • These findings provide insights into the structural dynamics and cellular integration of Thy-1.
