Area of Science:

  • Biochemistry
  • Enzymology
  • Proteomics

Background:

  • Mammalian calcium-activated proteases (calpains) play crucial roles in cellular processes.
  • Accurate monitoring of calpain activity is essential for understanding their function and associated diseases.
  • Existing methods may have limitations in sensitivity or specificity for calpain detection.

Purpose of the Study:

  • To highlight casein zymography as a gold standard assay for calpain detection.
  • To explain the underlying principles and advantages of the casein zymography technique.
  • To demonstrate its utility in studying calpain isoforms and inhibitor interactions.

Main Methods:

  • Utilizes casein as a substrate embedded within a gel matrix.
  • Calpain activity is detected by casein digestion, visualized as clear zones after Coomassie blue staining.

  • Employs native gel electrophoresis to separate calpain isoforms based on differential mobility.
  • Main Results:

    • Casein zymography effectively detects major calpain isoforms (Calpain-1, 2, and 3).
    • The method provides clear visualization of enzyme activity through specific substrate digestion.
    • Differential migration patterns allow for the identification of distinct calpain isoforms.

    Conclusions:

    • Casein zymography is a robust and reliable method for monitoring calpain activity in diverse biological samples.
    • Its ability to differentiate isoforms and assess inhibitor reversibility enhances its value in biochemical research.
    • This technique serves as a gold standard for calpain analysis.
