Background:

  • The Federal Bureau of Investigation (FBI) mandated an expansion of the CODIS core loci from 13 to 20.
  • This expansion aims to increase international compatibility and improve the efficiency of national DNA databases for law enforcement.

Purpose of the Study:

  • To evaluate and compare the performance of three new 6-dye STR amplification kits for forensic DNA profiling.
  • To determine the suitability of these kits for laboratories adopting the expanded CODIS core loci.

Main Methods:

  • Performance assessment of Investigator 24plex QS Kit (QIAGEN), GlobalFiler™ PCR Amplification Kit (Applied Biosystems™), and PowerPlex® Fusion 6C System (Promega).
  • Evaluation included sensitivity, inhibitor tolerance, heterozygous peak height, intra-color signal balance, minor component profiling in mixtures, and mixture ratio preservation.

Main Results:

  • GlobalFiler™ demonstrated the highest sensitivity.
  • Investigator 24plex QS Kit showed superior tolerance to PCR inhibitors (Humic acid, Tannic acid).
  • GlobalFiler™ and Fusion 6C provided better heterozygous peak height and intra-color signal balance. GlobalFiler™ and Investigator 24plex were better at profiling minor mixture components, while Fusion 6C maintained mixture ratios more consistently.

Conclusions:

  • The three evaluated STR amplification systems possess distinct performance characteristics.
  • Laboratories should select kits based on specific operational needs and requirements for forensic DNA analysis.
