    Summary

    Enphenamic acid (En.A.), a non-steroidal anti-inflammatory drug, alters blood pressure effects of clonidine. En.A. potentiated hypertension and blocked hypotension, suggesting a peripheral vascular interaction.

    Background:

    • Clonidine is an antihypertensive agent.
    • Non-steroidal anti-inflammatory drugs (NSAIDs) can affect cardiovascular parameters.

    Purpose of the Study:

    • To investigate the drug interaction between clonidine and enphenamic acid.
    • To elucidate the mechanism of this interaction on blood pressure regulation.

    Main Methods:

    • Drug interaction study in anesthetized dogs.
    • Administration of enphenamic acid prior to clonidine.
    • Intracerebroventricular administration of enphenamic acid.
    • Studies in reserpinized animals and with tolazoline blockade.

    Main Results:

    • Enphenamic acid administered 1 hour before clonidine potentiated the hypertensive response and blocked the hypotensive response.
    • Intracerebroventricular enphenamic acid did not alter the blood pressure response to clonidine.
    • The interaction was absent in reserpinized animals and partially blocked by tolazoline.

    Conclusions:

    • Enphenamic acid significantly interferes with the blood pressure effects of clonidine.
    • The interaction appears to be primarily peripheral and vascular in nature.
