This study introduces a novel nanoplatform combining gene therapy and photothermal therapy (PTT) for cancer treatment. The system provides fluorescence imaging guidance for enhanced oncotherapy and visual synergistic tumor treatment.
Area of Science:
- Biomedical Engineering
- Nanotechnology
- Oncology
Background:
- Gene therapy and photothermal therapy (PTT) are promising cancer treatments.
- Current combined therapies lack fluorescence imaging guidance, limiting clinical application.
- Tungsten oxide (WO) nanoparticles offer potential for PTT and imaging.
Purpose of the Study:
- To develop a novel nanoplatform for fluorescence imaging-guided, combined gene therapy and PTT.
- To evaluate the synergistic therapeutic effect of the nanoplatform in cancer treatment.
- To demonstrate the potential for visual synergistic tumor treatment using this multi-modal nanosystem.
Main Methods:
- Synthesized tungsten oxide (WO) nanoparticles using a template method.
- Constructed a mesoporous silica nanoparticle-based nanoplatform loaded with Bax gene, WO, and indocyanine green (ICG).
- Evaluated the nanoplatform's efficacy in vitro and in vivo using near-infrared (NIR) laser excitation and fluorescence imaging.
Main Results:
- The developed nanoplatform successfully integrated gene therapy, PTT, and fluorescence imaging.
- In vivo experiments demonstrated significant solid tumor regression under 808nm NIR light irradiation.
- The nanoplatform exhibited synergistic therapeutic effects, combining gene therapy and PTT.
Conclusions:
- The novel nanoplatform shows potential as a fluorescence imaging-guided multi-modal therapeutic nanosystem.
- This approach enables visual synergistic treatment for tumors.
- The combination of gene therapy and PTT with imaging guidance represents a promising strategy in oncotherapy.

Cancer Therapies
However, cancer treatments can pose several challenges, as therapies used to kill cancer cells are generally also toxic to normal cells. Moreover, cancer cells mutate rapidly and can develop resistance to chemical agents or radiation therapy. Besides, all types of cancer cells may not respond to the same therapy. Some cancer cells respond to one...
