In multiple sclerosis (MS), specific T cell subsets like CD4+ and CD8+ TEMRA cells are elevated. Natalizumab treatment impacts T cell populations, affecting CD49d and CD26 expression.
Area of Science:
- Immunology
- Neuroimmunology
- T cell biology
Background:
- Multiple Sclerosis (MS) is a chronic inflammatory disease of the central nervous system.
- T cell subsets, including naïve, central memory (CM), effector memory (EM), and terminally differentiated effector memory (TEMRA) cells, play critical roles in immune responses.
- The expression of CD49d and CD26 on T cells is associated with immune cell trafficking and function, but their specific roles in MS pathogenesis require further elucidation.
Purpose of the Study:
- To characterize naïve, CM, EM, and TEMRA CD4+ and CD8+ T cells and their expression of CD49d and CD26 in the peripheral blood of MS patients and healthy controls.
- To investigate the impact of natalizumab, a disease-modifying therapy for MS, on these T cell subsets and their marker expression.
Main Methods:
- Flow cytometry was used to analyze peripheral blood mononuclear cells (PBMCs).
- Characterization of T cell subsets (naïve, CM, EM, TEMRA) for both CD4+ and CD8+ T cells.
- Quantification of CD49d and CD26 expression on these T cell subsets.
Main Results:
- Increased frequencies of CD26+ CD28+ CD4+ TEMRA T cells were observed in all MS subtypes.
- Elevated levels of CD26+ CD28+ CD8+ TEMRA T cells were found in relapsing-remitting and secondary progressive MS.
- In progressive MS, CD49d+ CM T cells were decreased. Natalizumab treatment increased the overall number of circulating T cell subsets but decreased the frequency of most subsets expressing CD49d and CD26.
Conclusions:
- Specific T cell subsets, particularly CD4+ and CD8+ TEMRA cells expressing CD26, are altered in the peripheral blood of MS patients.
- CD49d+ CM T cells may be depleted in progressive MS.
- Natalizumab influences T cell distribution and marker expression, suggesting complex immunomodulatory effects in MS treatment.
