Background:

  • Traditional processing of Chinese medicinal herbs is common.
  • Accurate identification of processed herbs is challenging.
  • DNA mini-barcoding offers a novel approach for authentication.

Purpose of the Study:

  • To evaluate DNA mini-barcoding loci for identifying processed Chinese medicinal herbs.
  • To assess the amplification efficiency and identification accuracy of the trnL (UAA) intron P6 loop.

Main Methods:

  • Six DNA barcode loci (ITS2, psbA-trnH, rbcL, matK, trnL (UAA) intron, and its P6 loop) were used.
  • 45 processed samples from 15 species were analyzed.
  • Amplification rates and BLAST analysis against GenBank were performed.

Main Results:

  • The trnL (UAA) intron P6 loop showed a high amplification rate (75.56%).
  • Conventional loci had low amplification rates (8.89%-20%).
  • 45.23% of samples were identified to the genus level, with one to the species level.

Conclusions:

  • The trnL (UAA) intron P6 loop is a promising mini-barcode candidate for processed medicinal herb authentication.
  • It can serve as a complementary barcode for quality control.
  • This method will aid in medicinal materials control and marketplace integrity.
