DNA Microarrays02:34

DNA Microarrays

21.4K
Microarrays are high-throughput and relatively inexpensive assays that can be automated to analyze large quantities of data at a time. They are used in genome-wide studies to compare gene or protein expression under two varied conditions, such as healthy and diseased states. Microarrays consist of glass or silica slides on which probe molecules are covalently attached through surface functionalization. Most commonly, the slides are prepared through the chemisorption of silanes to silica...

Ribosome Profiling02:24

Ribosome Profiling

4.2K
Ribosome profiling or ribo-sequencing is a deep sequencing technique that produces a snapshot of active translation in a cell. It selectively sequences the mRNAs protected by ribosomes to get an insight into a cell’s translation landscape at any given point in time.
Applications of ribosome profiling
Ribosome profiling has many applications, including in vivo monitoring of translation inside a particular organ or tissue type and quantifying new protein synthesis levels.
The technique...

Summary

One-step reverse transcription loop-mediated isothermal amplification (RT-LAMP) offers a fast and sensitive method for human gene expression analysis. This technique is a viable alternative to reverse transcription polymerase chain reaction (RT-PCR), especially in resource-limited settings.

Area of Science:

  • Molecular Biology
  • Immunology
  • Biotechnology

Background:

  • Tumor Necrosis Factor-alpha (TNF-α) plays a key role in immune regulation and fever induction.
  • Reverse transcription polymerase chain reaction (RT-PCR) is a standard for gene expression analysis.
  • Loop-mediated isothermal amplification (LAMP) presents a promising alternative to PCR due to its specificity and sensitivity.

Purpose of the Study:

  • To compare the efficacy of one-step RT-PCR, two-step RT-LAMP, and one-step RT-LAMP for human gene expression analysis.
  • To evaluate these methods using the human TNF-α gene as a biomarker.

Main Methods:

  • Analysis of total RNA from peripheral blood cells of three febrile patients.
  • Comparative assessment of one-step RT-PCR, two-step RT-LAMP, and one-step RT-LAMP for gene expression.

Main Results:

  • One-step RT-PCR and one-step RT-LAMP demonstrated equivalent detection limits.
  • Two-step RT-LAMP exhibited a lower detection limit compared to the other methods.
  • One-step RT-LAMP was faster and yielded easier result interpretation.

Conclusions:

  • One-step RT-LAMP is a rapid, sensitive, and potentially valuable tool for gene expression analysis.
  • This method is suitable for environments with limited specialized equipment or expertise.
  • One-step RT-LAMP serves as a complementary technique to established methods like RT-PCR.
