Abstract:
The receptor tyrosine kinase KIT promotes survival and migration of melanocytes during development, and excessive KIT activity hyperactivates the RAS/MAPK pathway and can drive formation of melanomas, most notably of rare melanomas that occur on volar and mucosal surfaces of the skin. The much larger fraction of melanomas that occur on sun-exposed skin is driven primarily by BRAF- or NRAS-activating mutations, but these melanomas exhibit a surprising loss of KIT expression, which raises the question of whether loss of KIT in these tumors facilitates tumorigenesis. To address this question, we introduced a kit(lf) mutation into a strain of Tg(mitfa:BRAF melanoma-prone zebrafish. Melanoma onset was accelerated in kit(lf); Tg(mitfa:BRAF fish. Tumors from kit(lf) animals were more invasive and had higher RAS/MAPK pathway activation. KIT knockdown also increased RAS/MAPK pathway activation in a BRAFV600E-mutant human melanoma cell line. We found that pathway stimulation upstream of BRAFV600E could paradoxically reduce signaling downstream of BRAFV600E, and wild-type BRAF was necessary for this effect, suggesting that its activation can dampen oncogenic BRAFV600E signaling. In vivo, expression of wild-type BRAF delayed melanoma onset, but only in a kit-dependent manner. Together, these results suggest that KIT can activate signaling through wild-type RAF proteins, thus interfering with oncogenic BRAFV600E-driven melanoma formation. Insights
Loss of KIT receptor tyrosine kinase accelerates melanoma development and increases tumor invasiveness. KIT signaling through wild-type RAF proteins may interfere with BRAF-driven melanoma formation.
Area of Science:
- Oncology
- Molecular Biology
- Dermatology
Background:
- The receptor tyrosine kinase KIT is crucial for melanocyte development and survival.
- KIT hyperactivity drives melanoma formation, particularly rare types on volar and mucosal surfaces.
- Common melanomas on sun-exposed skin, driven by BRAF/NRAS mutations, paradoxically show reduced KIT expression.
Purpose of the Study:
- To investigate the role of KIT loss in the tumorigenesis of common melanomas.
- To determine if KIT loss facilitates melanoma development and progression.
- To elucidate the interaction between KIT signaling and the RAS/MAPK pathway in melanoma.
Main Methods:
- Introduction of a hypomorphic KIT mutation (kit(lf)) into a BRAF-driven zebrafish melanoma model.
- Analysis of melanoma onset, invasiveness, and RAS/MAPK pathway activation in genetically modified zebrafish.
- KIT knockdown experiments in BRAF V600E-mutant human melanoma cell lines.
- Investigation of wild-type BRAF's role in modulating oncogenic signaling.
Main Results:
- Melanoma onset was accelerated in kit(lf); Tg(mitfa:BRAF) zebrafish.
- Tumors from kit(lf) zebrafish exhibited increased invasiveness and elevated RAS/MAPK pathway activation.
- KIT knockdown in human melanoma cells also increased RAS/MAPK pathway activation.
- Wild-type BRAF was necessary to dampen oncogenic BRAF V600E signaling, a process dependent on KIT expression.
Conclusions:
- Loss of KIT expression can promote melanoma development and invasiveness.
- KIT signaling, potentially through wild-type RAF proteins, can act as a brake on oncogenic BRAF V600E-driven melanoma.
- These findings suggest a complex interplay between KIT and BRAF in melanoma pathogenesis.
