Summary
Genetic changes in PIK3CA and PTEN were investigated in Diffuse Large B-cell Lymphoma (DLBCL). PIK3CA amplification and PTEN deficiency were observed at the DNA level, suggesting roles in DLBCL pathogenesis.
Area of Science:
- Oncology
- Genetics
- Molecular Biology
Background:
- PIK3CA amplification and PTEN deficiency are known at the protein level in Diffuse Large B-cell Lymphoma (DLBCL).
- The clinical significance of PIK3CA and PTEN genetic alterations at the DNA level in DLBCL remains unestablished.
Purpose of the Study:
- To investigate the clinicopathological significance of PIK3CA and PTEN genetic status in DLBCL.
- To evaluate the association between genetic changes in PIK3CA and PTEN and clinicopathological variables in DLBCL patients.
Main Methods:
- Fluorescent in-situ hybridization (FISH) was used to assess PIK3CA and PTEN genetic changes in 205 DLBCL tissue samples.
- Cross-table analysis was employed to correlate genetic alterations with various clinicopathological variables.
Main Results:
- PIK3CA amplification occurred in 12.7% (26/205) and PTEN deficiency in 12.2% (25/205) of DLBCL cases at the DNA level.
- No significant associations were found between PIK3CA/PTEN genetic changes and clinicopathological variables.
- No significant correlation was observed between PTEN loss and PIK3CA amplification.
Conclusions:
- PTEN deficiency and PIK3CA amplification at the DNA level are events in the pathogenesis of DLBCL.
- These genetic alterations may play a role in DLBCL development, independent of clinicopathological factors.
