Gallbladder

2.4K
The gallbladder is a small, pear-shaped organ that plays a crucial role in our digestive system. Measuring about 10 cm in length, it is comparable in size to a kiwi fruit and is located in a hollow area on the lower surface of the liver. The gallbladder's primary function is to store and concentrate bile, a fluid produced by the liver that aids in digestion.
The gallbladder's anatomy consists of three regions: the fundus, body, and neck. Extending from the neck, the cystic duct joins...

Area of Science:

  • Pharmacology
  • Pharmaceutical Technology
  • Natural Product Chemistry

Background:

  • Gallnut (Galla chinensis) possesses medicinal properties, but its rectal administration requires a suitable dosage form.
  • Developing effective and safe rectal delivery systems is crucial for utilizing natural compounds.

Purpose of the Study:

  • To research the preparation process of gallnut suppositories using its water extract.
  • To evaluate the irritation potential of the gallnut suppositories on rectal mucosa.

Main Methods:

  • Gallnut extract prepared via decoction; suppositories formulated using fusion method with semi-synthetic aliphatic esters and rose flower oil.
  • Assayed suppository weight difference, in vitro melting time, and polyphenol, tannin, and saccharide content via UV-Vis spectrophotometry.
  • Evaluated colon mucosa irritation in New Zealand white rabbits after 14 days of successive administration.

Main Results:

  • Optimized prescription: semi-synthetic aliphatic esters and rose flower oil (2:1 ratio) as matrix, 54% drug loading.
  • Prepared suppositories: brown, conical, smooth; weight (1.43±0.03) g; melting time (17±2) min.
  • Contents per suppository: Polyphenols 332.4 mg•g-1, Tannic 245.0 mg•g-1, Polysaccharide 3.3 mg•g-1. No rectal irritation observed.

Conclusions:

  • The gallnut suppository is an acceptable dosage form with a controllable preparation process.
  • The developed suppositories demonstrated no irritation to the rectal mucosa in animal studies.
  • This formulation offers a safe and effective rectal administration route for gallnut extract.
