Response Surface Methodology01:16

Response Surface Methodology

685
Response Surface Methodology (RSM) is a collection of statistical and mathematical techniques used to develop, improve, and optimize processes. It is particularly valuable when many input variables or factors potentially influence a response variable.
The process of RSM involves several key steps:

Olefin Metathesis Polymerization: Ring-Opening Metathesis Polymerization (ROMP)01:16

Olefin Metathesis Polymerization: Ring-Opening Metathesis Polymerization (ROMP)

3.2K
Ring-opening metathesis polymerization or ROMP involves strained cycloalkenes as starting materials. The mechanism of ROMP proceeds by reacting cycloalkene with Grubbs catalyst to give metallacyclobutane intermediate which undergoes a ring-opening reaction to form new carbene. The new carbene reacts with another molecule of cycloalkene. Repetition of these steps leads to the formation of an unsaturated open-chain polymer product. All these steps are reversible, however, relieving the ring...

Actin Polymerization01:42

Actin Polymerization

8.7K
Actin polymerization occurs through the head-to-tail association of binding sites on monomeric actin or G-actin to form filamentous or F-actin. The polymerization can be divided into three phases ̶  nucleation, elongation, and steady-state phase.
The nucleation phase involves forming a stable nucleus consisting of three actin monomers to form a new actin filament. Actin-binding proteins such as formins and Arp2/3 complex help filament growth post-nucleation. The Formins form straight...

Optimal Foraging00:48

Optimal Foraging

14.0K
How animals obtain and eat their food is called foraging behavior. Foraging can include searching for plants and hunting for prey and depends on the species and environment.

Optimization Problems01:26

Optimization Problems

85
Optimization problems often involve identifying maximum or minimum values under specific constraints. A well-known example is determining the longest horizontal pipe that can be moved around a right-angled corner, where a 3-meter-wide hallway meets a 2-meter-wide hallway. This scenario, common in architectural design and industrial transport, can be understood conceptually through geometric and trigonometric reasoning. To visualize the problem, consider the pipe as a straight line that touches...

Impact Loading01:19

Impact Loading

730
Impact loading occurs when a moving object collides with a stationary structure, such as a rod with a uniform cross-sectional area fixed at one end. Under these conditions, the rod absorbs the kinetic energy from the striking object, leading to deformation and subsequent stress development. As the rod returns to its original position and reaches maximum stress, the absorbed energy, initially manifested as kinetic energy, transforms entirely into strain energy.
In cases of elastic deformation,...

Area of Science:

  • Pharmaceutical Sciences
  • Materials Science
  • Drug Delivery Systems

Background:

  • Aripiprazole (ARP) exhibits poor aqueous solubility, limiting its therapeutic efficacy.
  • Recent patents highlight the potential of Aripiprazole and Pluronic F127 micelles in pharmaceutical formulations.

Purpose of the Study:

  • To encapsulate Aripiprazole (ARP) within Pluronic F127 micelles to enhance its aqueous solubility.
  • To optimize the micellar formulation for improved drug delivery.

Main Methods:

  • Fabrication of Aripiprazole-loaded Pluronic F127 micelles using thin-film hydration.
  • Optimization of micellar formulation via response surface methodology (RSM).
  • Characterization using PDI, zeta potential, DLC, EE, in vitro drug release, DSC, FT-IR, P-XRD, TEM, and DLS.

Main Results:

  • Optimized conditions yielded micelles with PDI of 0.228, zeta potential of -4.04 mV, DLC of 76.50%, and EE of 18.56%.
  • In vitro studies demonstrated ~97% Aripiprazole release from micelles within 20h, thrice that of the pure drug.
  • TEM and DLS confirmed spherical micelles with a hydrodynamic diameter of 170.3 nm.

Conclusions:

  • The developed Pluronic F127 micellar system effectively enhances Aripiprazole solubility.
  • This polymeric micellar system shows significant potential as a drug delivery carrier for Aripiprazole.
