    Background:

    • Cholinergic systems are implicated in memory functions.
    • Evidence suggests differential roles in working and reference memory.

    Purpose of the Study:

    • To investigate the role of basocortical cholinergic neurons in memory.
    • To test if these neurons are differentially involved in working versus reference memory.

    Main Methods:

    • Unilateral injections of quinolinic acid or kainic acid into the nucleus basalis magnocellularis (nbm) in rats.
    • Behavioral testing using T-maze and radial arm maze tasks.
    • Biochemical assays of choline acetyltransferase (CAT) activity in cortical and hippocampal tissues.

    Main Results:

    • Quinolinate lesions impaired performance on a working memory task (T-maze).
    • Kainate lesions increased working memory errors more than reference memory errors in the radial arm maze.
    • Lesions reduced cortical CAT activity, with no significant change in hippocampal CAT activity.

    Conclusions:

    • Basocortical cholinergic neurons play a differential role in working and reference memory.
    • These findings support the hypothesis of specialized cholinergic system involvement in distinct memory types.
