Summary

Genetic polymorphisms in immunoglobulin G (IgG) Fc receptors (FcγR) are linked to generalized early-onset periodontitis (G-EOP) susceptibility in Japanese individuals. The FcγRIIIb-NA2 allele and FcγRIIIa-158F may increase the risk for G-EOP.

  • Previous research suggests a link between FcγR gene variations and periodontal disease severity.

Purpose of the Study:

  • To investigate the association between FcγR polymorphisms and generalized early-onset periodontitis (G-EOP) in a Japanese population.
  • To determine if specific FcγR genotypes contribute to G-EOP susceptibility.

Main Methods:

  • Case-control study involving 38 Japanese G-EOP patients, 83 AP patients, and 104 healthy controls.
  • FcγR genotypes for 3 bi-allelic polymorphisms were analyzed using allele-specific polymerase chain reactions.
  • Clinical parameters including probing depth, clinical attachment level, and alveolar bone level were assessed.

Main Results:

  • Significant differences in FcγRIIIb genotypes were observed between G-EOP patients and healthy controls (P=0.02).
  • The FcγRIIIb-NA2 allele was significantly over-represented in G-EOP patients compared to AP patients and controls (P=0.02, P=0.009).
  • A strong association was found between G-EOP and a composite genotype (FcγRIIIb-NA2 and FcγRIIIa-158F), with an odds ratio of 2.4 (P=0.04).

Conclusions:

  • The FcγRIIIb-NA2 allele is potentially associated with susceptibility to G-EOP in Japanese patients.
  • FcγRIIIa-158F may also play a role in G-EOP development.
  • These findings highlight the role of immunogenetic factors in the pathogenesis of early-onset periodontitis.
