Immune system dysregulation may contribute to Autism Spectrum Disorders (ASDs). This study found elevated levels of TNF-α, IL-6, and IL-17, and decreased IL-2 in ASD patients, supporting immune involvement in this neurodevelopmental condition.

Background:
- The exact causes of Autism Spectrum Disorders (ASDs), a group of severe neurodevelopmental conditions, remain unknown.

Purpose of the Study:
- To investigate the mRNA expression levels of key immune-related cytokines in individuals with ASD.
- To determine if specific cytokine expression patterns are associated with ASD.
Main Methods:
- Real-time quantitative polymerase chain reaction (qPCR) was used to measure the mRNA expression of ten cytokines (TNF-α, TGF-β, IFN-γ, CXCL8, IL-1β, IL-2, IL-4, IL-6, IL-17) in whole blood samples.
- The study included 30 patients diagnosed with ASD and 41 age- and sex-matched healthy control subjects.
Main Results:
- Significant upregulation of Tumor Necrosis Factor-alpha (TNF-α), Interleukin-6 (IL-6), and Interleukin-17 (IL-17) was observed in ASD patients compared to healthy controls (P < 0.0001, P = 0.001, P < 0.0001, respectively).
- A significant downregulation of Interleukin-2 (IL-2) was found in the ASD group (P < 0.0001).
- No significant differences in the expression levels of other tested cytokines were detected between the groups.
Conclusions:
- The findings provide further evidence supporting the hypothesis of immune system dysregulation in individuals with ASD.
- Altered expression of specific cytokines like TNF-α, IL-6, IL-17, and IL-2 may be implicated in the immune mechanisms underlying ASD.
