Four new dimeric acylphloroglucinols, hyperjaponicols A-D, were isolated from Hypericum japonicum. These compounds, along with others, demonstrated significant antibacterial activity and moderate lipase inhibition.
Area of Science:
- Natural Product Chemistry
- Medicinal Chemistry
- Pharmacognosy
Background:
- Hypericum japonicum is a plant species known to contain various bioactive compounds.
- Acylphloroglucinols are a class of natural products with diverse biological activities.
- Understanding the chemical constituents and bioactivities of medicinal plants is crucial for drug discovery.
Purpose of the Study:
- To isolate and characterize new dimeric acylphloroglucinols from Hypericum japonicum.
- To elucidate the structures of isolated compounds using spectroscopic methods.
- To evaluate the antibacterial and lipase inhibitory activities of the isolated compounds.
Main Methods:
- Isolation of compounds using chromatographic techniques.
- Structure elucidation through Nuclear Magnetic Resonance (NMR) and Mass Spectrometry (MS) data analysis.
- Single-crystal X-ray diffraction for determining relative configuration of known compounds.
- Determination of Minimum Inhibitory Concentration (MIC) for antibacterial activity.
- Measurement of lipase inhibitory activity using IC50 values.
Main Results:
- Four new dimeric acylphloroglucinols, named hyperjaponicols A-D (1-4), were successfully isolated.
- The structures of the new compounds were confirmed by extensive spectroscopic analysis.
- The relative configuration of sarothralen C (5) was reassigned using NMR and X-ray diffraction.
- Compounds 1-4, 6, and 7 displayed significant antibacterial activity against four bacterial strains (MIC: 0.8-3.4 μM).
- Compound 3 exhibited moderate lipase inhibitory activity (IC50: 8.3 μM).
Conclusions:
- Hypericum japonicum is a rich source of novel dimeric acylphloroglucinols.
- The isolated hyperjaponicols A-D possess significant antibacterial properties.
- This study contributes to the understanding of the chemical diversity and pharmacological potential of Hypericum species.

Nucleosomes, paradoxically, perform two opposite functions simultaneously. On the one hand, their main responsibility is to protect the delicate DNA strands from physical damage and help achieve a higher compaction ratio. While on the other hand, they must allow polymerase enzymes to access DNA...
