    Background:

    • Genistein, a phytoestrogen, possesses both cytotoxic and chemopreventive properties.
    • Its cytoprotective and antioxidant effects warrant investigation in cancer research.

    Purpose of the Study:

    • To evaluate the cytotoxic, cytoprotective, and antioxidant properties of genistein.
    • To compare genistein's effects with cisplatin and beta-estradiol on cancer and normal cells.

    Main Methods:

    • Spectrophotometric MTT assay was used to assess cytotoxicity.
    • Effects on mitochondrial pore induction were evaluated using rat liver mitochondria.

    Main Results:

    • Genistein demonstrated cytotoxic effects only at high concentrations (IC50 = 20 mM for RDF, 14 mM for HeLa cells).
    • Genistein exhibited cytoprotective and antioxidant properties at physiological concentrations (up to 200 nM).
    • Unlike cisplatin, genistein did not induce mitochondrial pore formation.

    Conclusions:

    • Genistein acts as a cytoprotective agent at physiological concentrations.
    • Its significant antioxidant activity suggests potential as a chemopreventive drug.
    • Genistein's differential effects on cancer and normal cells warrant further study.
