Summary

Interleukin gene polymorphisms, specifically IL-1beta(-511C/T) in Europeans and IL-1beta(+3954C/T) in Americans, are linked to an increased risk of recurrent aphthous stomatitis (RAS). However, IL-10(-1082G/A) polymorphism shows no association with RAS risk.

Background:

  • Recurrent aphthous stomatitis (RAS) is a common oral mucosal disease with multifactorial etiology.
  • Genetic factors, particularly polymorphisms in cytokine genes, are implicated in RAS susceptibility.

Purpose of the Study:

  • To conduct a meta-analysis assessing the association between specific interleukin gene polymorphisms (IL-1beta, IL-6, IL-10) and the risk of recurrent aphthous stomatitis (RAS).

Main Methods:

  • A systematic literature search was performed on PubMed and Embase databases.
  • A meta-analysis of 10 studies involving 884 cases and 1104 controls was conducted using Revman 5.3 software.
  • Pooled odds ratios (OR) and 95% confidence intervals (CI) were calculated to evaluate the relationship between polymorphisms and RAS risk.

Main Results:

  • The IL-1beta(-511C/T) polymorphism significantly increased RAS risk in Europeans (e.g., T vs. C: OR=1.35, 95%CI=1.09-1.67).
  • The IL-1beta(+3954C/T) C allele was associated with increased RAS risk in Americans (C vs. T: OR=1.52, 95%CI=1.07-2.17).
  • No significant association was found between IL-10(-1082G/A) polymorphism and RAS risk in Americans.

Conclusions:

  • The IL-1beta(-511C/T) and IL-1beta(+3954C/T) gene polymorphisms are associated with an increased risk of recurrent aphthous stomatitis in European and American populations, respectively.
  • The IL-10(-1082G/A) polymorphism does not appear to be associated with RAS risk in Americans.
  • Further research is warranted to validate these findings and explore underlying mechanisms.
