We developed a novel steroidal prodrug supramolecular hydrogel using ionic coordination. This hydrogel shows potential as an ophthalmic drug delivery system for anti-inflammatory therapy with tunable drug release and low cytotoxicity.
Area of Science:
- Materials Science
- Drug Delivery
- Biomedical Engineering
Background:
- Steroidal prodrugs offer therapeutic benefits but require effective delivery systems.
- Supramolecular hydrogels provide a versatile platform for controlled drug release.
- Ionic coordination is an emerging strategy for hydrogel design.
Purpose of the Study:
- To design a steroidal prodrug supramolecular hydrogel using ionic coordination.
- To investigate the properties and drug release behavior of the hydrogel.
- To evaluate the potential of the hydrogel as an ophthalmic drug delivery system for anti-inflammatory therapy.
Main Methods:
- Formation of supramolecular hydrogels via ionic coordination of dexamethasone succinate (Dex-SA) with divalent cations (e.g., Ca2+).
- Rheological analysis to determine mechanical and thixotropic properties.
- In vitro drug release studies and stability assessments.
- Cytotoxicity assays and anti-inflammatory efficacy evaluation in macrophage models.
Main Results:
- The Ca2+/Dex-SA supramolecular hydrogel exhibited elastic and thixotropic properties.
- Tunable, gradual release of dexamethasone and Dex-SA was achieved, influenced by Ca2+ concentration.
- The hydrogel showed good storage stability as a xerogel and was easily rehydrated.
- Negligible cytotoxicity and comparable anti-inflammatory efficacy to dexamethasone were observed.
Conclusions:
- Cation-instructed steroidal prodrug supramolecular hydrogels are feasible.
- This hydrogel system demonstrates promise for ophthalmic anti-inflammatory drug delivery.
- The tunable drug release and favorable biocompatibility support its potential clinical application.
