    Background:

    • Serological studies revealed cross-reactivity between Human Leukocyte Antigen (HLA)-B17 and HLA-A locus products.
    • A specific epitope, defined by monoclonal antibody MA2.1, is shared by HLA-A2 and HLA-B17 subtypes (Bw57, Bw58).
    • Understanding the structural basis of these serological relationships is crucial for HLA research.

    Purpose of the Study:

    • To investigate the structural basis of cross-reactivity between HLA-Bw58 and HLA-A locus molecules.
    • To determine the nucleotide sequence of the HLA-Bw58 gene and compare its protein product with other HLA molecules.
    • To identify unique sequence features within the HLA-Bw58 gene.

    Main Methods:

    • Isolation of the HLA-Bw58 gene from the WT49 B cell line.
    • Transfection of the HLA-Bw58 gene into mouse L cells for protein expression and characterization.
    • Determination of the nucleotide sequence of the HLA-Bw58 gene (3520 bp) and comparison of deduced protein sequences with other HLA-A,B,C molecules.

    Main Results:

    • The HLA-Bw58 alpha 1 domain exhibits unusual homology to both HLA-A and HLA-B locus products.
    • Specific amino acid and nucleotide sequence homologies were identified between HLA-Bw58 and HLA-A2/Aw24 in the polymorphic alpha 1 domain.
    • Novel inverted repeat sequences were discovered in the HLA-Bw58 gene, flanking splice sites for alpha 2 and alpha 3 exons.

    Conclusions:

    • Structural similarities in the alpha 1 domain provide a basis for the observed serological cross-reactivity between HLA-Bw58 and HLA-A molecules.
    • The identified sequence homologies, particularly around residues 62-65, may be involved in the MA2.1 epitope.
    • The unique sequence features in HLA-Bw58 suggest distinct evolutionary or regulatory mechanisms compared to other HLA-A,B,C genes.
