Purpose of the Study:

  • To identify medications with potential to improve treatment outcomes for individuals dependent on amphetamines.

Main Methods:

  • A comprehensive literature search was conducted across major databases (Medline, PsycINFO, Embase, Cochrane) from 1997 to 2016.
  • Studies on pharmacotherapies for amphetamine/methamphetamine dependence were selected and assessed.
  • A meta-narrative review approach was employed to synthesize findings from 49 eligible studies, including 33 randomized controlled trials (RCTs).

Main Results:

  • Twenty potential pharmacotherapies were investigated, with five showing limited evidence for reducing amphetamine use: methylphenidate, bupropion, modafinil, and naltrexone.
  • Dexamphetamine showed benefits in treatment retention but not in reducing amphetamine use.
  • Six other medications indicated potential efficacy, but insufficient studies preclude definitive conclusions.

Conclusions:

  • No single medication currently demonstrates sufficient and consistent evidence for routine treatment of amphetamine use disorder.
  • High rates of study dropout and poor medication adherence present significant challenges to evidence strength and clinical application.
  • Further research is crucial to identify effective strategies for improving patient engagement and treatment outcomes in pharmacotherapy for amphetamine dependence.
