Total internal reflection fluorescence microscopy or TIRF is an advanced microscopic technique used to visualize fluorophores in samples close to a solid surface with a higher refractive index, such as a glass coverslip. Summary

A short LED-fluorescence microscopy (FM) training course is insufficient for laboratory technicians. However, practical "in situ" training significantly improves their ability to accurately detect bacilli, especially with sufficient daily slide volume.

  • Technicians examined slides stained with Ziehl Neelsen (ZN) and FM, progressing to blinded testing.

  • Effective skill acquisition in LED-FM is possible through a short "in situ" phase, even without on-site expert support.
  • Training effectiveness is linked to a minimum workload, with 750 slides/year recommended.
