Area of Science:

  • Animal Science
  • Microbiology
  • Gastroenterology

Background:

  • Weaning compromises intestinal barrier function and increases infection risk in piglets.
  • Probiotics can improve gut health by reinforcing the intestinal epithelium and modulating gut microbiota.
  • Mechanisms of probiotic action on intestinal epithelium and microbiota require further elucidation.

Purpose of the Study:

  • To investigate the protective effects of Lactobacillus plantarum ZLP001 on the piglet intestinal epithelium and microbiota.
  • To determine how modulated microbiota by L. plantarum ZLP001 influences epithelial function.

Main Methods:

  • Assessed paracellular permeability using fluorescein isothiocyanate-dextran (FD-4).
  • Measured tight junction (TJ) proteins, proinflammatory cytokines, and host defense peptides via RT-qPCR, ELISA, and western blot.

  • Analyzed short-chain fatty acid (SCFA) concentrations and fecal microbiota composition using ion chromatography and high-throughput sequencing.
  • Main Results:

    • L. plantarum ZLP001 counteracted enterotoxigenic Escherichia coli (ETEC)-induced gut permeability and TJ protein reduction.
    • The probiotic downregulated proinflammatory cytokines (IL-6, IL-8, TNFα) and increased host defense peptides (pBD2, PG1-5).
    • L. plantarum ZLP001 increased beneficial butyrate-producing bacteria (Anaerotruncus, Faecalibacterium) and Lactobacillus, while decreasing Clostridium sensu stricto 1.

    Conclusions:

    • L. plantarum ZLP001 enhances intestinal barrier function in piglets.
    • The probiotic strengthens epithelial defense mechanisms and modulates gut microbiota composition.
    • These actions contribute to improved gut health and reduced infection risk in weaned piglets.
