The long noncoding RNA NEAT1 promotes osteosarcoma by sponging miR-339-5p, leading to increased TGF-β1 expression and tumor progression. This NEAT1/miR-339-5p/TGF-β1 pathway is a potential therapeutic target for osteosarcoma.
Area of Science:
- Molecular Biology
- Oncology
- Genetics
Background:
- The long noncoding RNA (lncRNA) nuclear enriched abundant transcript 1 (NEAT1) is implicated in various cancers.
- The specific role and molecular mechanisms of NEAT1 in osteosarcoma (OS) remain largely uncharacterized.
Purpose of the Study:
- To investigate the function of NEAT1 in osteosarcoma development.
- To elucidate the molecular mechanism underlying NEAT1's role in OS progression.
- To identify potential therapeutic targets for osteosarcoma.
Main Methods:
- Quantitative reverse-transcription polymerase chain reaction (qRT-PCR) to assess NEAT1 expression.
- Lentivirus-mediated downregulation of NEAT1 to evaluate its effects on OS cell behavior.
- MiRNA sponge assays to determine interactions between NEAT1, miR-339-5p, and TGF-β1.
Main Results:
- NEAT1 expression was detected in OS cell lines.
- Inhibition of NEAT1 significantly suppressed OS cell proliferation, migration, and invasion.
- NEAT1 was found to sponge miR-339-5p, and miR-339-5p overexpression repressed OS proliferation.
- NEAT1 promotes OS by sponging miR-339-5p, leading to increased transforming growth factor β1 (TGF-β1) expression.
Conclusions:
- NEAT1 acts as an oncogene in osteosarcoma by inhibiting miR-339-5p.
- The NEAT1/miR-339-5p/TGF-β1 axis plays a crucial role in osteosarcoma progression.
- This axis represents a potential novel molecular pathway and therapeutic target for osteosarcoma.
