Overview of DNA Repair02:25

Overview of DNA Repair

33.7K
In order to be passed through generations, genomic DNA must be undamaged and error-free. However, every day, DNA in a cell undergoes several thousand to a million damaging events by natural causes and external factors. Ionizing radiation such as UV rays, free radicals produced during cellular respiration, and hydrolytic damage from metabolic reactions can alter the structure of DNA. Damages caused include single-base alteration, base dimerization, chain breaks, and cross-linkage.
Chemically...

Quantitative Analysis01:12

Quantitative Analysis

1.4K
Quantitative analysis is a technique for measuring the amount of specific constituents in a sample. When the sample's composition is unknown, qualitative analysis is performed first to identify its components, which ensures that the correct substances are measured during the quantitative phase.
In quantitative analysis, two key measurements are made: the sample quantity and a property proportional to the amount of the analyte (the substance being analyzed). This forms the basis of the...

Translesion DNA Polymerases02:10

Translesion DNA Polymerases

11.2K
Translesion (TLS) polymerases rescue stalled DNA polymerases at sites of damaged bases by replacing the replicative polymerase and installing a nucleotide across the damaged site. Doing so, TLS allows additional time for the cell to repair the damage before resuming regular DNA replication.
TLS polymerases are found in all three domains of life - archaea, bacteria, and eukaryotes. Of the different classes of TLS polymerases, members of the Y family are fitted with specialized structures that...

Cancer Survival Analysis01:21

Cancer Survival Analysis

765
Cancer survival analysis focuses on quantifying and interpreting the time from a key starting point, such as diagnosis or the initiation of treatment, to a specific endpoint, such as remission or death. This analysis provides critical insights into treatment effectiveness and factors that influence patient outcomes, helping to shape clinical decisions and guide prognostic evaluations. A cornerstone of oncology research, survival analysis tackles the challenges of skewed, non-normally...

    Area of Science:

    • Biomedical imaging
    • Cytopathology
    • Medical diagnostics

    Background:

    • Traditional cervical cancer screening methods, including The Bethesda System (TBS) and DNA quantitative analysis, have limitations in sensitivity, specificity, and efficiency.
    • Current combined screening approaches require separate cell smears, proving time-consuming and inconvenient.
    • A need exists for an integrated screening method that combines TBS classification and DNA quantitative analysis on a single sample.

    Purpose of the Study:

    • To develop and validate a novel cervical cancer screening method integrating TBS classification and DNA quantitative analysis on the same cell smear.
    • To overcome the limitations of existing methods, including low sensitivity, poor specificity, and procedural inconvenience.
    • To improve the accuracy and efficiency of early cervical cancer detection.

    Main Methods:

    • Developed a method for simultaneous Pap and Feulgen staining on a single cell smear to enable dual analysis.
    • Established a multispectral imaging system and a DNA absorbance peeling model based on linear multiple regression to accurately quantify DNA absorbance.
    • Synthesized pseudo-color images from 3-band cell images for TBS classification, integrating it with DNA quantitative analysis.

    Main Results:

    • The developed DNA quantitative analysis model demonstrated stability and minimal error.
    • The integrated method achieved a high diagnostic rate for cervical cancer screening.
    • Pseudo-color images generated for TBS screening were bright and clear, facilitating accurate classification.

    Conclusions:

    • The proposed method successfully integrates TBS classification and DNA quantitative analysis on a single cell smear, overcoming previous limitations.
    • This innovative approach offers a stable, accurate, and efficient tool for cervical cancer diagnosis and early screening.
    • The method holds significant potential for improving cervical cancer detection rates and patient outcomes.
