This study identified 19 novel genes, including SOX4, associated with pancreatic cancer (PC) using transcriptome-wide association study (TWAS). These findings offer new insights into the genetic mechanisms underlying PC development.
Area of Science:
- Genetics
- Oncology
- Bioinformatics
Background:
- Pancreatic cancer (PC) remains a significant health challenge with complex genetic underpinnings.
- Identifying novel genetic factors is crucial for understanding PC pathogenesis and developing targeted therapies.
Purpose of the Study:
- To identify novel candidate genes associated with pancreatic cancer (PC).
- To explore the genetic architecture of PC through a comprehensive transcriptome-wide association study (TWAS).
Main Methods:
- Performed a transcriptome-wide association study (TWAS) using published PC GWAS summary data.
- Analyzed GWAS data involving 558,542 SNPs from 1896 PC cases and 1939 controls.
- Utilized FUSION software for tissue-specific TWAS and validated gene relevance with Oncomine, STRING, and CluePedia tools.
Main Results:
- Identified 19 genes significantly associated with PC, including LRP5L, SOX4, and EGLN3.
- KEGG pathway analysis revealed significant enrichment in pathways like 'One carbon pool by folate' and 'Cell cycle'.
- SOX4 was identified as an overlapping gene with previously reported overexpressed genes in PC patients.
Conclusions:
- Identified novel candidate genes and pathways implicated in pancreatic cancer.
- The findings provide new clues for investigating the genetic mechanisms of PC.
- This research contributes to a deeper understanding of PC's genetic basis.
