Preoperative treatment with Tetrahydrocurcumin (THC) improves islet function and reduces apoptosis before transplantation. This antioxidant may enhance outcomes for islet transplantation in diabetes treatment.

Purpose of the Study:

  • To determine if preoperative Tetrahydrocurcumin (THC) treatment can reduce ischemic damage and apoptosis in pancreatic islets.
  • To evaluate the impact of THC on islet viability and function in vitro and after cytokine exposure.

Main Methods:

  • Islets were cultured with or without THC.
  • In vitro islet viability and function were assessed.
  • Islets were exposed to a cytokine cocktail, and apoptosis-related proteins were analyzed in INS-1 cells.

Main Results:

  • THC-treated islets showed 1.3-fold higher glucose-induced insulin secretion.
  • THC improved glucose-induced insulin release and nitric oxide (NO) production after cytokine challenge.
  • Apoptosis was significantly reduced, and B-cell lymphoma-2 (an anti-apoptotic protein) was elevated in THC-treated islets.

Conclusions:

  • Preoperative THC administration enhances islet function and viability.
  • THC attenuates cytokine-induced damage and apoptosis, suggesting a protective role in islet transplantation.
