New Genes for Leaf Rust Resistance in CIMMYT Durum Wheats.

Plant disease·2019

Researchers identified new wheat genes for resistance to leaf and stripe rust diseases. These findings, particularly a novel locus in the Chilero wheat line, can improve breeding strategies for disease-resistant crops.

Area of Science:

  • Plant Genetics
  • Agricultural Science
  • Pathology

Background:

  • The 'Chilero' spring wheat line exhibits significant resistance to leaf rust and stripe rust.
  • Understanding the genetic basis of this resistance is crucial for developing durable disease management strategies in wheat.

Purpose of the Study:

  • To identify and map genes conferring resistance to leaf rust and stripe rust in a recombinant inbred line (RIL) population derived from Avocet × Chilero.
  • To characterize the genetic architecture of rust resistance in wheat.

Main Methods:

  • Field trials were conducted across three Mexican locations from 2012-2015 to assess rust resistance.
  • Genotyping was performed using DArT-array, DArT-GBS, and SSR markers, with 6,168 polymorphic markers used for genetic mapping.

Related Experiment Videos

  • Inclusive composite interval mapping was employed to detect resistance loci.
  • Main Results:

    • Four colocated resistance loci for both rust diseases and two stripe rust resistance loci were identified.
    • A known pleotropic gene (Lr46/Yr29) on chromosome 1BL and a novel colocated resistance locus (QLr.cim-5DS/QYr.cim-5DS) in Chilero were confirmed.
    • A new stripe rust resistance locus on chromosome 7BL and two minor QTLs from Avocet on chromosomes 1DL and 4BS were mapped.

    Conclusions:

    • The study successfully mapped multiple quantitative trait loci (QTLs) for leaf and stripe rust resistance in wheat.
    • The identified resistance loci, including a novel one in Chilero, provide valuable targets for marker-assisted wheat breeding.
    • Flanking SNP markers can be developed into KASP markers for practical application in breeding programs.
