Dermal mesenchymal stem cells (DMSCs) from psoriasis patients show altered gene expression, potentially driving immune cell migration in this chronic inflammatory disease. These findings suggest DMSCs play a key role in psoriasis pathogenesis.
Area of Science:
- Immunology
- Dermatology
- Stem Cell Biology
Background:
- Psoriasis is a chronic, systemic, immune-mediated inflammatory disease.
- Mesenchymal stem cells modulate the inflammatory microenvironment by influencing immunocyte behavior.
Purpose of the Study:
- To investigate the role of dermal mesenchymal stem cells (DMSCs) in immune cell migration in psoriasis patients.
- To analyze the expression of migration-associated genes in DMSCs from psoriasis patients.
Main Methods:
- Recruited 22 psoriasis patients and 22 healthy controls.
- Analyzed seven migration-associated genes (CMKLR-1, COL8A-1, NETO-2, NRK, SFRP, SULF-2, SYTL-2) using qPCR and Western blot.
- Assessed peripheral blood-derived mononuclear cell (PBMC) migration to DMSCs using a Transwell chamber system.
Main Results:
- Upregulation of CMKLR-1, COL8A-1, NETO-2, NRK, SYTL-2, and SULF-2 mRNA and protein in psoriatic DMSCs.
- Significant downregulation of SFRP-2 in psoriatic DMSCs.
- Increased in vitro migration of normal PBMCs towards psoriatic DMSCs (6.3-fold increase).
Conclusions:
- DMSCs may be pathologically involved in monocyte migration in psoriasis.
- DMSCs might regulate immune responses and be central to the "psoriatic march."
- Altered gene expression in DMSCs contributes to psoriasis pathogenesis.
