Tonsil-derived mesenchymal stem cells (MSCs) modulate T-lymphocyte responses. These MSCs suppress T-cell proliferation and shift cytokine production towards an anti-inflammatory profile.
Area of Science:
- Immunology
- Regenerative Medicine
- Stem Cell Biology
Background:
- Mesenchymal stem cells (MSCs) possess immunomodulatory properties.
- Tonsils are a readily accessible source of MSCs in children.
- Understanding MSC immunomodulation is crucial for therapeutic applications.
Purpose of the Study:
- To evaluate the immunomodulatory effects of tonsil-derived MSCs.
- To assess the impact of MSCs on T-lymphocyte proliferation.
- To determine the influence of MSCs on T helper 1 (Th1) and T helper 2 (Th2) cytokine production.
Main Methods:
- Tonsils were collected from pediatric patients undergoing tonsillectomy.
- Tonsil-derived MSCs were co-cultured with peripheral blood mononuclear cells (PBMCs).
- PBMCs were stimulated with phytohemagglutinin (PHA) to induce proliferation and differentiation into Th1 or Th2 cells.
Main Results:
- Tonsil-derived MSCs significantly suppressed PHA-induced PBMC proliferation.
- Co-culture with MSCs led to a significant decrease in interferon-gamma (IFN-γ) production (Th1 cytokine).
- Co-culture with MSCs resulted in a significant increase in interleukin-4 (IL-4) production (Th2 cytokine).
Conclusions:
- Tonsil-derived MSCs exhibit potent immunomodulatory capabilities.
- These MSCs effectively suppress T-lymphocyte proliferation.
- Tonsil-derived MSCs promote a shift in T-cell cytokine profiles, favoring Th2 over Th1 responses.
