    Background:

    • The cornea, an avascular eye tissue, is exposed to high concentrations of ascorbic acid in the aqueous humor.
    • Lipoxygenase products, such as 12-hydroxyeicosatetraenoic acid (12-HETE), are generated by the cornea.
    • The role of ascorbic acid in modulating corneal lipoxygenase activity is not well understood.

    Purpose of the Study:

    • To investigate the influence of ascorbic acid on lipoxygenase product generation in the rabbit cornea.
    • To determine if ascorbic acid's antioxidant properties mediate its effect on corneal lipoxygenase.

    Main Methods:

    • Rabbit corneal homogenates were incubated with [14C]arachidonic acid.
    • Lipoxygenase metabolites were separated using thin-layer chromatography.
    • Metabolite quantification was performed using liquid scintillation counting.

    Main Results:

    • Physiological concentrations of ascorbic acid significantly inhibited the formation of 12-HETE, a major lipoxygenase product.
    • Ascorbic acid did not alter prostaglandin E2 (PGE2) formation.
    • Inhibition of 12-HETE formation by ascorbic acid was observed even in the presence of catalase, ruling out hydrogen peroxide as the primary inhibitory agent.
    • High concentrations of hydrogen peroxide and another antioxidant, 2,6-dichlorophenol-indophenol, also inhibited 12-HETE formation.

    Conclusions:

    • Ascorbic acid inhibits corneal lipoxygenase activity, likely through its antioxidant properties.
    • The findings suggest that ascorbic acid in the aqueous humor may modulate lipoxygenase activity in the cornea.
