Functional magnetic resonance imaging (fMRI) connectivity reliably measures brain coupling. This finding supports using fMRI biomarkers to predict neuropsychiatric disorder risk and treatment response.

Purpose of the Study:

  • To assess the reliability of task-modulated amygdala connectivity with the dorsomedial frontal/cingulate cortex (DMFC).
  • To evaluate the potential of these connectivity measures as reliable biomarkers for clinical applications.

Main Methods:

  • Assessed task-modulated coupling between amygdala/subgenual anterior cingulate cortex and DMFC using psychophysiological interaction analysis.
  • Scanned 29 healthy individuals twice, approximately two weeks apart.
  • Used three emotional face-processing tasks (identification, matching, gender classification) with two runs per session.

Main Results:

  • Found good-to-excellent within-subject reliability for amygdala-DMFC coupling.
  • Reliability was demonstrated both between days (in two tasks) and within a day (in one task).

Conclusions:

  • Task-modulated amygdala-DMFC coupling exhibits good reliability.
  • These findings suggest that regional coupling measures are reliable enough for future clinical studies predicting treatment response or risk.
