Abstract

Insights

Colony-stimulating factor 1 receptor (Csf1r) and Mer tyrosine kinases regulate Kupffer cell function. Inhibiting Csf1r or Mer in mice impairs liver regeneration and reduces Kupffer cell cytokine production.

Area of Science:

  • Immunology
  • Hepatology
  • Macrophage Biology

Background:

  • Kupffer cells (KCs) are tissue-resident macrophages in the liver.
  • KCs exist as CD11bhi and F4/80hi subsets.
  • Tyrosine kinases Csf1r and Mer are expressed on tissue-resident macrophages, but their role in KCs and liver regeneration is unclear.

Purpose of the Study:

  • To investigate the expression of Csf1r and Mer on murine KC subsets.
  • To determine the role of Csf1r and Mer in liver regeneration (LR).

Main Methods:

  • Flow cytometry to characterize KC subsets from wild-type and Csf1r-GFP mice.
  • Partial hepatectomy (PH) in mice treated with Csf1r or Mer inhibitors, or clodronate liposomes.
  • Analysis of serum and liver tissues; in vitro testing of sorted KC subsets.

Main Results:

  • Mer is expressed on F4/80hi KCs, with 55% co-expressing Csf1r.
  • Csf1r inhibition reduced F4/80hi KCs by 50% and delayed LR by 20%.
  • Csf1r inhibition increased liver injury and decreased proliferation post-PH; Mer inhibition had milder effects.

Conclusions:

  • F4/80hi KCs are Mer+, with Csf1r expression defining a subset.
  • Csf1r and Mer inhibition reduce KC cytokine production.
  • Inhibition of Csf1r or Mer delays liver regeneration.

Liver Regeneration01:24

Liver Regeneration

The liver is an important organ in vertebrates that plays an essential role in metabolism. It is also responsible for storing and redistributing nutrients such as carbohydrates, fats, and vitamins in the body. Additionally, the liver releases bile salts which are critical for digesting food and eliminating toxic metabolites from the body.
Cells of Liver
The liver comprises four major types of cells— hepatocytes, stellate, Kupffer, and sinusoidal endothelial cells. The hepatocytes are...

Feedback Inhibition00:46

Feedback Inhibition

Biochemical reactions are occurring constantly in cells, converting starting substances to different products, usually with the help of enzymes that speed the reactions. Without enzymes, it would take far too long for most reactions to occur to be useful to the cell!

Stem Cell Therapy for Tissue Regeneration01:21

Stem Cell Therapy for Tissue Regeneration

Stem cell therapy is a method used in regenerative medicine to repair and restore function to damaged tissues and organs. Stem cells have the potential to proliferate and differentiate into various tissue types, making them ideal candidates for tissue regeneration. For example, hematopoietic stem cell transplants are commonly used in blood cancer treatment to replenish damaged bone marrow and restore healthy blood cells.

Enzyme Inhibition01:30

Enzyme Inhibition

Inhibitors are molecules that reduce enzyme activity by binding to the enzyme. In a normally functioning cell, enzymes are regulated by a variety of inhibitors. Drugs and other toxins can also inhibit enzymes. Some inhibitors bind to the enzyme’s active site, while others inhibit enzymatic activity by binding to other sites on the protein structure.

Inhibition of Cdk Activity02:34

Inhibition of Cdk Activity

The orderly progression of the cell cycle depends on the activation of Cdk protein by binding to its cyclin partner. However, the cell cycle must be restricted when undergoing abnormal changes. Most cancers correlate to the deregulated cell cycle, and since Cdks are a central component of the cell cycle, Cdk inhibitors are extensively studied to develop anticancer agents. For instance, cyclin D associates with several Cdks, such as Cdk 4/6, to form an active complex. The cyclin D-Cdk4/6 complex...

Overview of Regeneration and Repair01:19

Overview of Regeneration and Repair

Regeneration and repair processes are critical in healing damages caused by injury, disease, and aging. In regeneration, the damaged tissue is entirely replaced with new growth that restores the original architecture and function. In contrast, tissue repair usually results in a fixed tissue architecture involving scar formation. Scars generally do not reestablish tissue function and may also exhibit structural abnormalities at the injury site.
