Background:

  • Effective cancer vaccines require optimal targeting of nanoparticles (NPs) to dendritic cells (DCs) for antigen delivery.
  • Dendritic cell-specific intercellular adhesion molecule-3 grabbing non-integrin (DC-SIGN) is a key receptor on DCs for immune modulation.

Purpose of the Study:

  • To compare the efficacy of antibody-targeted versus natural ligand (ICAM3-Fc)-targeted NPs for DC activation and T cell response.
  • To evaluate the impact of NP targeting strategies on the induction of anti-tumour immunity.

Main Methods:

  • Poly (lactic-co-glycolic acid) (PLGA) NPs were formulated with tetanus toxoid, gp100 antigen, and toll-like receptor adjuvants.
  • NPs were targeted to DC-SIGN on DCs using either specific humanized antibodies or ICAM3-Fc fusion proteins.
  • DC activation markers (CD80, CD83, CD86, CCR7) and T cell activation via cross-presentation were analyzed.

Main Results:

  • Antibody-targeted NPs showed higher binding and uptake by DCs compared to ICAM3-Fc targeted NPs.
  • No significant differences were observed in DC activation markers between the two targeting groups.
  • DCs loaded with ICAM3-Fc targeted NPs demonstrated enhanced potency in T cell activation through cross-presentation.

Conclusions:

  • Targeting NPs with natural ligands like ICAM3-Fc may be more effective for inducing T cell-mediated anti-tumour immunity than using antibodies.
  • The choice of targeting strategy for NP vaccines significantly impacts their immunogenicity and potential efficacy in cancer therapy.
